چکیده
The effects of partially purified thrombopoietin (TPO). prepared from the plasma of thrombocytopenic rabbits, upon megakaryocyte colony-forming cells (Meg-CFC), megakaryocytes, and platelets were evaluated. The thrombopoiesis-stimulating activity of both types of preparations of TPO used was established by their ability to increase the levels of selenomethionine-75Se (75SeM) in the circulating platelets of mice, as described previously. TPO did not increase the frequency or total numbers of Meg-CFC or granulocyte-macrophage-CFC (GM-CFC) in the bone marrow or spleen of recipient mice. Similarly, the addition of TPO directly to 0.3% agar cultures of bone marrow or splenic cells did not alter the frequency of either Meg-CFC or GM-CFC, although in some experiments. concentrations greater than required to stimulate thrombopoiesis in vivo were used. TPO did not enhance the ability of suboptimal concentrations of spleen conditioned medium to support colony growth. Human erythropoietin (EPO) at concentrations from 0.6 U/mI to 8 U/mI, in vitro, did not increase the frequency of bone marrow or splenic Meg-CFC or GM-CFC. As previously reported, two types of megakaryocyte

Thrombopoietin (c-mpl Ligand) Acts Synergistically With Erythropoietin, Stem Cell Factor, and Interleukin-l1 to Enhance Murine Megakaryocyte Colony Growth and Increases Megakaryocyte Ploidy In Vitro
Thrombopoietin (Tpo), the ligand for the ompl receptor, is a major regulator of platelet production in vivo. Treatment of mice with purified recombinant Tpo increases platelet count fourfold and expands colony-forming unit-megakaryocyte (CFU-Meg) numbers. Other cytokines including interleukin-3 (IL-3). IL-6, IL-11, erythropoietin (Epo), and stem cell factor (SCF) can stimulate megakaryopoiesis....

Thrombopoietin (also called c-Mpl ligand, megakaryocyte growth and development factor, megapoietin) has recently been purified and cloned. This molecule is indeed the long-sought-after hematopoietic factor that controls platelet production. Thrombopoietin levels increase within 24 h after the onset of thrombocytopenia and are inversely and exponentially proportional to the platelet count. Injec...
متن کاملA single injection of pegylated murine megakaryocyte growth and development factor (MGDF) into mice is sufficient to produce a profound stimulation of megakaryocyte frequency, size, and ploidization.
Despite numerous studies investigating the action of c-mpl ligand, no reports have defined the in vivo changes in megakaryocytopoiesis in response to a single injection of this cytokine. Here we compare the kinetics of the megakaryocytopoietic response in C57BI/6J mice administered 25 micrograms/ kg or 250 micrograms/kg of pegylated (PEG) murine megakaryocyte growth and development factor (MGDF...
متن کاملThrombopoietin: biology and potential clinical applications.
After an almost 40-year search for a primary regulatory of platelet production, thrombopoietin has recently been purified and cloned. Thrombopoietin regulates all stages in the production of platelets by promoting both the proliferation of megakaryocyte progenitors and their maturation into platelet-producing megakaryocytes. In preclinical studies in normal mice and non-human primates, administ...
